- Take Osmolex ER exactly as prescribed by your doctor. The starting dosage is 129 mg by mouth once daily in the morning. It may be increased to 322 mg by mouth once daily. Never break, crush, chew, or split Osmolex ER tablets, as it will break the time-release mechanism.
